Abstract

Structure-guided drug design led to the identification of a class of spirocyclic ureas which potently inhibit human 11β-HSD1 in vitro. Lead compound 10j was shown to be orally bioavailable in three species, distributed into adipose tissue in the mouse, and its (R) isomer 10j2 was efficacious in a primate pharmacodynamic model.

    • Accelerated skin wound healing by selective 11β-Hydroxylase (CYP11B1) inhibitors

2018, European Journal of Medicinal Chemistry
      Citation Excerpt :

Another study in mice skin showed an increase of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1, reduces 11-dehydrocorticosterone to corticosterone in rodents) expression, but a lack of CYP11B1 expression during wound healing [10]. Blockade of 11β-HSD1 by subcutaneous application of the inhibitor 2 (IC50 11β-HSD1 = 1.1 nM, Fig. 2) [13] resulted in enhanced wound healing in mice (in vivo) [14]. In fact, 1 is a non-selective inhibitor of CYP enzymes [15] and, in our studies, inhibits 11β-HSD1 (54% at 50 μM and 70% at 200 μM, Fig. 2) as well.

A number of adamantine-containing selective 11β-HSD1 inhibitors have been reported [96]. Tice et al. used them to generate models of the 11β-HSD1 binding site with the help of a proprietary structure-based drug design program of Vitae Pharmaceuticals called Contour [97]. The most satisfactory poses were selected, and based on them a medicinal chemistry program was initiated to improve potency, selectivity, and physical properties, supported by additional modeling.

However, due to their nature, each of the established stereoselective methods has certain limits in substrate scope and a complementary stereoselective synthetic route toward varied 3-benzylpiperidines might be of high value. 3-Acylacrylic acids have proven to be very good substrates for crystallization-induced asymmetric transformations (CIAT), yielding a variety of functionalized α-amino acids in excellent yields and stereoselectivities.11–13 We envisioned that 4-aroylpent-4-enoic acids 9 in conjunction with aza-Michael additions might be utilized in a similar fashion, since they have structural features characteristic for known successful CIAT systems (Scheme 1).
